Aflatoxin B1 Rapid Test Kit
1. Principle
This kit is based on competitive indirect immunochromatography technology, in which aflatoxin B1 in sample competes for the colloid gold labeled antibody with aflatoxin B1 coupling antigen captured on test line. The test result can be watched by naked eye.
2. Application
This kit is developed for rapid test of aflatoxin B1 contamination in grain, feed and edible oil.

5. Sample preparation
5.1 Grain
5.1.1 Take 3.0±0.05g grinded sample into a 50ml centrifuge tube.
5.1.2 Add 4ml of 80% methanol, cover tightly, vortex for 5min, and then centrifuge for separation: room temperature / 5min / 3000g.
5.1.3 Dilute the solution according to the chart based on your requirement

5.2 Feed
5.2.1 Take 3.0±0.05g of grinded sample into a 50ml centrifuge tube.
5.2.2 Add 6ml of 80% methanol, cover tightly, vortex for 5min, and then centrifuge for separation: room temperature / 5min / 3000g.
5.2.3 Dilute the solution according to the chart based on your requirement

5.3 Edible oil
5.4.1 Take 3.0±0.05g grinded sample into a 15ml centrifuge tube.
5.4.2 Add 3ml of 80% methanol, cover tightly, vortex for 5min, and then centrifuge for separation: room temperature / 5min / 3000g.
5.4.3 Take 1ml of the upper lever clear solution into a 5ml centrifuge tube, add 1ml of n-hexane, cover tightly, shake upside down fiercely for 10 times, then centrifuge for separation: room temperature / 3min / 3000g.
5.4.4 Take 100μl of the down level solution into a 500μl centrifuge tube, add 200μl of sample buffer, mix completely and ready to use
NOTE: Please adjust the pH to 7 with 1M NaOH / HCl if the pH of the prepared solution without the range of 6-8.
6. Assay procedure
6.1 Read the instructions carefully before experiment. Bring the test kit and samples to room temperature.
6.2 Take bottles needed from the kit package, take out required wells and strips, and make proper marks. Please use these test strips within 1h. Seal the cap of the bottles. The rest strips can be stored for future use.
6.3 Take 200ul of the test samples into the wells, then repeatedly absorb and drop for 5 times to mix the sample with the reagent in the wells completely. The mixture should be pink, and then start the timer.
6.4 Incubate for 5min at ambient temperature; insert the test strips into the wells with the "Sample End" end fully dipped in to the mixture.
6.5 Incubate for 5min at ambient temperature again. Take out the strip; judge the result according to Part 7.
7. Result interpretation
Negative (-): Test Line and Control Line are both red: indicating that aflatoxin B1 in sample is lower than the LOD of the kit.
Positive (+): No color shows in Test line, indicating that aflatoxin B1 in sample is higher than the LOD of the kit.
Invalid: No color shows in Control Line, indicating the operation is incorrect or the test kit is out of date. In this case, please read the instruction again carefully, and do the assay again with new strips.
